Root carbohydrate storage in young saplings of an Amazonian tidal várzea forest before the onset of the wet season Acta Botanica
Scarano,Fabio Rubio; Cattânio,José Henrique; Crawford,Robert M.M..
Root starch and glucose content were measured for young saplings of 18 Amazonian tidal várzea tree species during a dry season. The pattern of carbohydrate storage depended on the type of plant involved and soil topography which is directly linked to flood regime. Most plants showed high root carbohydrate content at this point in the dry season, however, several typically flood-tolerant species (particularly palm trees) presented a low root carbohydrate content, suggesting a strategy of acquiring reserves during the wet season to survive the dry season, when depletion occurs. Plant survival in these flood-prone forests seems to be the result of more than only one adaptive mechanism.

Proteolytic release and partial characterization of human sperm-surface glycopeptides BJMBR
Sperm-surface glycopeptides were obtained from intact sperm membranes after proteolytic release by different enzymatic treatments such as autoproteolysis, trypsin, papain and pronase. Glycopeptides were isolated, their properties and composition were examined, and their monosaccharide and amino acid constituents were characterized. The monosaccharides identified were fucose, mannose, galactose, N-acetylglucosamine, and N-acetylgalactosamine, which form part of more than one type of oligosaccharide units. Autoproteolytic treatment mainly provided O-glycosidic type oligosaccharides, while a mixture of O- and N-glycosidic oligosaccharides was obtained in variable proportions when treated with trypsin, papain or pronase. The highest degree of peptide cleavage...

Intracellular and intercellular compartmentation of carbohydrate metabolism in leaves of temperate gramineae Rev. Bras. Bot.
POLLOCK,CHRISTOPHER J.; FARRAR,JOHN; KOROLEVA,OLGA A.; GALLAGHER,JOE A.; TOMOS,A. DERI.
This review considers the importance of compartmentation in the regulation of carbohydrate metabolism in leaves. We draw particular attention to the role of the vacuole as a site for storage of soluble sugars based on sucrose, and discuss briefly their characteristic metabolism. We also point out inconsistencies between the observed properties of vacuoles and the behaviour in vitro of the enzymes of fructan biosynthesis that do not support the hypothesis that the vacuole is the site of synthesis as well as of storage. We also consider compartmentation of carbohydrate metabolism between different cell types, using mainly our studies on leaves of temperate C3 gramineae. Here we present evidence of significant differences in carbon metabolism between...

Ferulic acid depletion by cultured soybean seedlings under action of glucose and methionine BABT
Herrig,Vanessa; Silva,Hélio Cândido da; Ferrarese,Maria de Lourdes Lucio; Ferrarese-Filho,Osvaldo.
Cultured soybean seedlings were used to investigate how glucose or methionine influenced depletion of ferulic acid. Three-day-old seedlings were grown in hydroponic solution containing ferulic acid plus glucose or methionine, and the level of the phenolic acid were monitored in the nutrient culture. The results showed that ferulic acid depletion was more rapid in the presence of those compounds. After 6 h, the increase caused by glucose (0.01 and 0.05 mM) was more pronounced than methionine in the same concentrations. On the other hand, methionine (0.1 and 0.2 mM) increased depletion more significantly than glucose. Results suggested that both compounds might to increase the allelopathic effects of ferulic acid in the seedlings.

Pyruvate kinase activity as index of carbohydrate assimilation by mesozooplankton: an early field implementation in the Bay of Biscay, NE Atlantic ArchiMer
Bergeron, Jean-pierre; Herbland, Alain.
Estimates of carbohydrate assimilation within the mesozooplankton compartment are a fundamental concern in oceanography because they give a general expression of the biologically-regulated incorporation of metabolic fuel produced from inorganic nutrients by phytoplankton in the sea. A new tool is proposed for the assessment of such processes. It is based on the measurement of the activity of an enzyme, pyruvate kinase (PK), on samples of the whole mesozooplankton community. Here, the results are presented of an early attempt at such an implementation in the Bay of Biscay (north-east Atlantic) in springtime. A main area situated in front of an estuary (Gironde) has been extensively studied-the same sampling grid was covered on three occasions (two 20 day...

Alteração no potencial osmótico e teor de carboidratos solúveis em plantas jovens de lobeira (Solanum lycocarpum St.-Hil.) em resposta ao estresse hídrico Rev. Bras. Bot.
CHAVES FILHO,JALES TEIXEIRA; STACCIARINI-SERAPHIN,ELIANE.

In vitro Dendrobium nobile plant growth and rooting in different sucrose concentrations Horticultura Brasileira
Faria,Ricardo T. de; Rodrigues,Fabiana N.; Oliveira,Luciana do V.R.; Müller,Cláudio.
Sucrose is a very important component in in vitro culture media, serving as a source of carbon and energy. In this paper, the rooting and in vitro growth of Dendrobium nobile Lindl (Orchidaceae) were studied using different sucrose concentrations (0 g L-1; 5 g L-1; 10 g L-1; 20 g L-1; 30 g L-1 and 60 g L-1), in a modified MS medium containing half the regular concentration of macronutrients at pH 5.8. Greater increases in plant height (4.21±0.6 cm) and high seedling multiplication (1:4) were observed in the 60 g L-1 sucrose treatment, even without the addition of plant hormones. Sucrose concentration in the culture medium did not influence in vitro plant rooting.

The alga Bryothamnion seaforthii contains carbohydrates with antinociceptive activity BJMBR
Vieira,L.A.P.; Freitas,A.L.P.; Feitosa,J.P.A.; Silva,D.C.; Viana,G.S.B..
Bryothamnion seaforthii, a red alga common to the Northeastern coast of Brazil, was used to prepare the protein fraction F0/60 by ammonium sulfate precipitation. The chromatography of F0/60 on DEAE-Sephadel column resulted in two lectin fractions, PI and PII, which have antinociceptive properties in rodents. We determined the antinociceptive activity of the PII fraction and of a carbohydrate-containing fraction (CF) in mice. The CF was prepared from the dried algae, after digestion with 100 mM sodium acetate, pH 6.0, containing 5 mM cysteine, EDTA and 0.4% papain, at 60ºC. A 10% cetylpyridinium chloride was added to the filtrate, and the precipitate was dissolved with 2 M NaCl:ethanol (100:15, v/v) followed by the carbohydrate precipitation with ethanol....

Effect of carbon source on morphology and histodifferentiation of Araucaria angustifolia embryogenic cultures BABT
Steiner,Neusa; Vieira,Felipe do Nascimento; Maldonado,Sara; Guerra,Miguel Pedro.
The aim of the present work was to establish in vitro conditions for the induction, stabilization and proliferation of embryogenic cultures of A. angustifolia. Pre-cotyledonary staged zygotic embryos inoculated BM medium supplemented with 5 µM 2,4- D, 2 µM BAP and Kin, and 3% maltose or sucrose resulted in 66.7% induction rate. The rate of induction of embryogenic cultures was affected by the carbon source, as well the multiplication and morphology of the embryogenic cultures. Embryogenic cultures maintained in BM medium with maltose presented bipolar morphology. Globular somatic embryos were obtained BM medium with 9% (PEG) and (9%) maltose. These results could establish an in vitro regenerative protocol towards the conservation and improvement of this...

Effect of sediment nearness on the metabolic enzyme activity and energy state of the oyster Crassostrea gigas ArchiMer
Le Moullac, Gilles; Fleury, Pierre-gildas; Le Coz, Jean-rene; Moal, Jeanne; Samain, Jean-francois.
This study was designed to assess in situ the effects of sediment nearness on Crassostrea gigas metabolism. One year-old oysters were reared for 5 months (April-August) in plastic bags on metallic frames at 10 and 60 cm off the sediment at the beginning of April. The management of the energetic resources (storage, consumption) was estimated with respect to biochemical changes of proteins, lipids and carbohydrates. In order to evaluate the effect of the sediment nearness at cellular level, the metabolic rate was evaluated by assessing pyruvate kinase (PK) and phosphoenolpyruvate carboxykinase (PEPCK) enzyme activities and electron transport system (ETS) activity. The metabolic pathways were assessed by measuring ATP and intermediary metabolites such as...

Methodological challenges in carbohydrate analyses R. Bras. Zootec.
Hall,Mary Beth.
Carbohydrates can provide up to 80% of the dry matter in animal diets, yet their specific evaluation for research and diet formulation is only now becoming a focus in the animal sciences. Partitioning of dietary carbohydrates for nutritional purposes should reflect differences in digestion and fermentation characteristics and effects on animal performance. Key challenges to designating nutritionally important carbohydrate fractions include classifying the carbohydrates in terms of nutritional characteristics, and selecting analytical methods that describe the desired fraction. The relative lack of information on digestion characteristics of various carbohydrates and their interactions with other fractions in diets means that fractions will not soon be...

Selection of artificial hosts for oviposition by wild Anastrepha obliqua (Macquart) (Diptera, Tephritidae): influence of adult food and effect of experience Rev. Bras. entomol.
Leal,Thamara A. B. S.; Zucoloto,Fernando S..
Several factors influence the selection of oviposition substrates by insects. The aim of the present work was to find answers to the following questions related to the oviposition behavior of Anastrepha obliqua. Can carbohydrates (glucose or sucrose) present in the adult diet have influence on the female preference for an oviposition substrate with similar composition? Can the previous experience with a host containing one of mentioned carbohydrates interfere in further selection of oviposition substrates? The results showed that the kind of carbohydrate present in the adult diet did not affect the female preference for an artificial oviposition substrate, neither when it was presented by itself nor in combination with brewer's yeast. The effect of...

Lectin-binding properties of Aeromonas caviae strains BJM
Rocha-de-Souza,Cláudio M.; Hirata-Jr,Raphael; Mattos-Guaraldi,Ana L.; Freitas-Almeida,Angela C.; Andrade,Arnaldo F. B..
The cell surface carbohydrates of four strains of Aeromonas caviae were analyzed by agglutination and lectin-binding assays employing twenty highly purified lectins encompassing all sugar specificities. With the exception of L-fucose and sialic acid, the sugar residues were detected in A. caviae strains. A marked difference, however, in the pattern of cell surface carbohydrates in different A. caviae isolates was observed. Specific receptors for Tritricum vulgaris (WGA), Lycopersicon esculentum (LEL) and Solanum tuberosum (STA) (D-GlcNAc-binding lectins) were found only in ATCC 15468 strain, whereas Euonymus europaeus (EEL, D-Gal-binding lectin) sites were present exclusively in AeQ32 strain, those for Helix pomatia (HPA, D-GalNAc-binding lectin) in AeC398...

Assessment of biochemical methods to detect enzymatic depolymerization of polysaccharides ArchiMer
Rigouin, Coraline; Delbarre Ladrat, Christine; Sinquin, Corinne; Colliec-jouault, Sylvia; Dion, M.
To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Reducing sugars assays are quantitative, sensitive and almost usable with all kind of polysaccharide but some compounds may interfere with them. When polysaccharide is...
